Using fluorescence microscopy for studying the preparation and stability of DNA SAMs with attention to the underlying surface crystallography

Abstract: Modifying surfaces for use as DNA sensors requires careful control over the DNA coverage, local environment and uniformity. The use of in-situ fluorescence microscopy provides a unique view of these surfaces, and with the appropriate substrate, characteristics of the DNA SAM can be obtained. Examples will be given to show that this in-situ surface analysis approach can be used to direct the methodology for making DNA SAMs, addressing the issues of optimizing coverage, uniformity, and stability.
